ABSTRACT
Title: Isolation, Characterization, and Assessment of the Bioremediation Potential of Lead (Pb)-Tolerant Bacteria from Batam Coastal Waters
Background: Lead (Pb) contamination in coastal waters is persistent and accumulative in the food chain, posing serious health hazards (neurological, renal, hematological effects, cancer). Batam City, an industrial coastal area, has reported Pb levels exceeding quality standards in water, sediment, and biota. Heavy-metal-degrading bacteria offer a cost-effective bioremediation approach. This study aimed to isolate Pb-tolerant bacteria from the coastal waters of Batam and evaluate their Pb-removal capability.
Method: Water and sediment samples were collected from Tanjung Uncang, Sekupang Port, and Pulau Sarang. Isolation was conducted using Marine Agar supplemented with 10 ppm Pb, followed by tolerance testing at Pb concentrations ranging from 50–1000 ppm using the turbidimetric method (OD600). The five best isolates were further evaluated for their Pb-removal ability using Atomic Absorption Spectrophotometry (AAS) and characterized for morphological and biochemical properties. All experiments were conducted in triplicate. Pb removal data were analyzed using one-way analysis of variance (ANOVA), followed by Tukey's Honestly Significant Difference (HSD) test at a 95% confidence level (α = 0.05).
Result: The physicochemical characteristics of the sampling sites showed temperatures of 28–29°C, pH 7.5–7.9, salinity 29–30‰, and dissolved oxygen 7.8–8.2 mg/L. A total of 32 Pb-tolerant bacterial isolates were successfully obtained, with sediment samples yielding more isolates than water samples. Six isolates survived at Pb concentrations up to 800 ppm, whereas none exhibited growth at 1000 ppm. Isolates TJS 3.6.2 and TJS 2.6.3 demonstrated the highest Pb reduction efficiencies, reaching 86,85% and 84,56%, respectively. Characterization results revealed the predominance of Gram-negative bacilli and coccobacilli, along with one Gram-positive coccus isolate.
Conclusion: Isolates TJS 3.6.2 and TJS 2.6.3 exhibited the highest potential as Pb bioremediation agents, as they tolerated Pb concentrations of up to 800 ppm and achieved Pb removal efficiencies of 86.85% and 84.56%, respectively. These findings indicate that sediments from the coastal waters of Batam are a promising source of indigenous bacteria for the development of heavy metal bioremediation technologies in coastal environments. These isolates are recommended for further studies involving molecular identification, optimization of bioremediation conditions, and field-scale applications.
