Uji Aktivitas Isolat L-Asparaginase Dari Rimpang Temulawak (Curcuma xanthorrhiza Roxb) Terhadap Sel Hela

Abstract
L-asparaginase adalah enzim hidrolase yang mengkatalisis hidrolisis asparagin menjadi asam aspartat dan amonia. L-asparaginase diketahui membantu pengobatan kanker dengan menghambat sintetis protein sel kanker tanpa merusak sel normal. Metode yang dilakukan adalah isolasi enzim meliputi ekstraksi, fraksinasi dengan ammonium sulfat; karakterisasi dengan uji aktivitas spesifik dan uji sitotoksisitas pada sel HeLa. Aktivitas spesifik tertinggi isolat L-asparaginase fraksi amonium sulfat terdapat pada fraksi keempat sebesar 22,639 unit/mg. Selanjutnya fraksi keempat diujikan pada sel HeLa dalam uji sitotoksisitas. Berdasarkan hasil uji sitotoksisitas, isolat ini memiliki aktivitas penghambatan terhadap pertumbuhan sel HeLa dengan nilai LC50 sebesar 309,74 ppm
