Penentuan kadar protein menurut metode lowry dilakukan dengan cara membuat larutan standar dengan berbagai tingkat konsentrasi, kemudian dari masing-masing konsentrasi tersebut diambil 1 mL dan ditambahkan larutan lowry B, kemudian dibiarkan selama 10 menit dan kemudian ditambahkan larutan lowry A untuk dibiarkan selama 20 menit lalu diukur absorbansinya pada panjang gelombang 600 nm. Hasil penelitian pengukuran absorbansi menggunakan spectronic 20D+ menunjukkan nilai R2 = 0,9927 sedangkan untuk Spectrophotometer uv-vis T 60U menunjukan nilai R2 = 0,9971.
