Abstract
Telah dilakukan karakterisasi enzim penisilin asilase hasil isolasi dari Eschericia coli B-130. Karakterisasi meliputi pH optimum, temperatur optimum, tetapan Michaelis Menten (Km), kecepatan maksimum (Vm). Aktivitas enzim penisilin asilase ditentukan dengan metode Kornfeld pada λ 538 nm. Hasil penelitian menunjukkan bahwa pH optimum = 7,5, temperatur optimum 55°C, tetapan Michaelis Menten (Km) = 0,86 μM kecepatan reaksi maksimum (Vm) untuk pembentukan produk 6-APA sebesar 147 μmol/mL enzim B-130
