Uji Aktivitas Antioksidan Senyawa Flavonoid dari Ekstrak Etanol Daun Johar (Senna siamea Lamk)

Abstract
Telah dilakukan penelitian tentang isolasi senyawa flavonoid dari ekstrak etanol daun johar (Senna siamea Lamk) serta uji aktivitas antioksidan. Penelitian ini bertujuan untuk memperoleh isolat flavonoid, mengidentifikasi dan menguji aktivitas antioksidan isolat flavonoid dari ekstrak etanol daun johar. Metode penelitian dibagi menjadi empat tahap, tahap pertama yaitu isolasi senyawa flavonoid. Tahap kedua yaitu pemisahan senyawa flavonoid menggunakan kromatografi cair vakum, kromatografi kolom gravitasi dan kromatografi lapis tipis preparatif. Tahap ketiga yaitu identifikasi isolat flavonoid menggunakan spektrofotometer UV-Vis dan FTIR. Tahap keempat adalah uji aktivitas antioksidan dengan metode DPPH. Hasil isolasi flavonoid didapatkan isolat flavonoid seberat 0,006 gram (0,004%). Identifikasi isolat flavonoid dengan menggunakan spektrofotometer UV-Vis menunjukkan bahwa isolat flavonoid memiliki struktur dasar senyawa flavon dengan serapan maksimum pada panjang gelombang pita I (350 nm) dan pita II (255 nm dan 267 nm) dan analisis menggunakan FTIR menunjukkan isolat flavonoid memiliki gugus fungsi O-H, C-H alifatik, C=O, C=C aromatik, C-O eter dan C-O alkohol sehingga diduga isolat flavonoid memiliki struktur dasar senyawa luteolin yang tersubstitusi. Hasil uji aktivitas antioksidan isolat flavonoid menunjukkan IC50 sebesar 139,8373 mg/L.
