Aspek biologi suatu spesies sangat penting diketahui untuk memahami sejarah hidup suatu spesies. Informasi ini penting sebagai dasar pengelolaan sumber daya tersebut di perairan. Ikan sembilang, Plotosus canius (Hamilton, 1822) telah dilaporkan menjadi sumber daya dengan status terancam di beberapa lokasi. Tujuan penelitian adalah untuk menganalisis aspek biologi ikan sembilang (Plotosidae: Siluriformes) di perairan Teluk Kolono yang mewakili tiga musim yaitu musim barat, musim peralihan barat ke timur, dan musim timur. Pengumpulan contoh ikan dilakukan setiap bulan dari bulan Februari hingga Agustus 2020 dengan bottom gillnet bermata jaring 1¼, 1¾, 2, dan 3 inci. P. canius yang tertangkap mempunyai kisaran panjang dan bobot berturut-turut 15,5–31,3 cm dan 25,3–253,8 g. Ikan jantan dan betina menunjukkan pola pertumbuhan isometrik (b=3,06) dan allometrik negatif (b=2,15), sementara faktor kondisi tertinggi ditemukan saat musim barat berturut-turut 1,18±0,12 dan 1,27±0,35. Rasio kelamin relatif seimbang yaitu 1:1, sementara indeks kematangan gonad (IKG) bervariasi. IKG jantan tertinggi sebesar 0,56±0,01 terjadi saat musim peralihan barat ke timur sedangkan betina sebesar 1,83±2,06 saat musim timur. Hasil temuan ini akan mendukung upaya pengelolaan dan pengembangan konservasi ikan sembilang secara efektif di masa datang.
