L-asparaginase adalah enzim hidrolase yang mengkatalisis hidrolisis asparagin menjadi asam aspartat dan amonia. Penelitian ini dilakukan melalui beberapa tahapan utama yaitu isolasi enzim, uji aktivitas dan uji potensi antikanker. Isolasi L-asparaginase melalui dua tahap yaitu ekstraksi dan fraksinasi bertingkat menggunakan garam amonium sulfat sehingga akan dihasilkan enzim L-asparaginase yang murni dari protein. Uji aktivitas spesifik L-asparaginase dilakukan pada kondisi optimum yaitu pada suhu 37 0C, pH 8,5, dan waktu inkubasi 30 menit dengan cara menghitung jumlah amonia yang terbentuk dengan pereaksi Nessler dan menghitung kadar protein dengan metode Lowry dari masing-masing fraksi. Dari hasil penelitian diperoleh L-asparaginase dari rimpang kunyit putih (Curcuma mangga Vall) dengan aktivitas spesifik yang besar bila dibandingkan dari sumber yang lain seperti benalu, bawang merah, bawang putih dan bawang bombay. Aktivitas spesifik enzim L-asparaginase kunyit putih sebesar 2195,715 unit/mg protein pada fraksi kelima (80-100%). Tahapan yang ketiga yaitu uji potensi antikanker pada kultur sel Leukimia tipe K562 dengan menggunakan metode viabilitas sel dengan penambahan MTT dan diukur dengan ELISA Reader. Pengujian dilakukan dengan 15 variasi konsentrasi dan diketahui bahwa enzim L-asparaginase mempunyai potensi yang baik sebagai antikanker dengan nilai LC50 pada konsentrasi penambahan L-asparaginase sebesar 9,419 ppm.}, issn = {2597-9914}, pages = {57--62} doi = {10.14710/jksa.11.3.57-62}, url = {https://ejournal.undip.ac.id/index.php/ksa/article/view/15097} }
